Karyotypic description of four species of Trigona (Jurine, 1807) (Hymenoptera, Apidae, Meliponini) from the State of Mato Grosso, Brazil Genet. Mol. Biol.
Costa,Kleber França; Brito,Rute Magalhães; Miyazawa,Carlos Suetoshi.
The genus Trigona contains at least 31 species, but there have been few cytogenetic studies of this group. In this work, four species of Trigona (T. branneri, T. chanchamayoensis, T. hyalinata, and T. recursa) from the municipality of Cuiabá, in the State of Mato Grosso, Brazil, were studied. In all of the species, the females had 2n = 34 chromosomes and the males had n = 17. The C-banding patterns showed that the karyotypes of these species consisted mainly of acrocentric and pseudoacrocentric chromosomes. These cytogenetic findings should useful in future phylogenetic studies of this group.

Karyotype characterization of Trigona fulviventris Guérin, 1835 (Hymenoptera, Meliponini) by C banding and fluorochrome staining: report of a new chromosome number in the genus Genet. Mol. Biol.
Domingues,Alayne Magalhães Trindade; Waldschmidt,Ana Maria; Andrade,Sintia Emmanuelle; Andrade-Souza,Vanderly; Alves,Rogério Marco de Oliveira; Silva Junior,Juvenal Cordeiro da; Costa,Marco Antônio.
Although many species of the genus Trigona have been taxonomically described, cytogenetic studies of these species are still rare. The aim of the present study was to obtain cytogenetic data by conventional staining, C banding and fluorochrome staining for the karyotype characterization of the species Trigona fulviventris. Cytogenetic analysis revealed that this species possesses a diploid chromosome number of 2n = 32, different from most other species of this genus studied so far. This variation was probably due to the centric fusion in a higher numbered ancestral karyotype, this fusion producing the large metacentric chromosome pair and the lower chromosome number observed in Trigona fulviventris. Heterochromatin was detected in the pericentromeric...

Cytogenetic characterization of Partamona cupira (Hymenoptera, Apidae) by fluorochromes Genet. Mol. Biol.
Marthe,Jefferson de Brito; Pompolo,Silvia das Graças; Campos,Lucio Antônio de Oliveira; Salomão,Tânia Maria Fernandes; Tavares,Mara Garcia.
Four colonies of the stingless bee Partamona cupira (Hymenoptera: Apidae) were cytogenetically analyzed using conventional staining and the fluorochromes CMA3 e DAPI. The females have 2n = 34 chromosomes (2K=32+2). Some females, however, presented an additional large B acrocentric chromosome, to a total of 2n = 35. Chromosome B and the chromosomal pairs 2, 9 and 10 showed CMA3+ bands, indicating an excess of CG base-pairs. A clear association was verified between the P. helleri B chromosome SCAR marker and the presence of a B chromosome in P....

Insights into the Melipona scutellaris (Hymenoptera, Apidae, Meliponini) fat body transcriptome Genet. Mol. Biol.
Sousa,Cristina Soares de; Serrão,José Eduardo; Bonetti,Ana Maria; Amaral,Isabel Marques Rodrigues; Kerr,Warwick Estevam; Maranhão,Andréa Queiroz; Ueira-Vieira,Carlos.
The insect fat body is a multifunctional organ analogous to the vertebrate liver. The fat body is involved in the metabolism of juvenile hormone, regulation of environmental stress, production of immunity regulator-like proteins in cells and protein storage. However, very little is known about the molecular mechanisms involved in fat body physiology in stingless bees. In this study, we analyzed the transcriptome of the fat body from the stingless bee Melipona scutellaris. In silico analysis of a set of cDNA library sequences yielded 1728 expressed sequence tags (ESTs) and 997 high-quality sequences that were assembled into 29 contigs and 117 singlets. The BLAST X tool showed that 86% of the ESTs shared similarity with Apis mellifera (honeybee) genes. The...

Comparative study on the use of specific and heterologous microsatellite primers in the stingless bees Melipona rufiventris and M. mondury (Hymenoptera, Apidae) Genet. Mol. Biol.
Lopes,Denilce Meneses; Campos,Lúcio Antônio de Oliveira; Salomão,Tânia Maria Fernandes; Tavares,Mara Garcia.
Due to their high degree of polymorphism, microsatellites are considered useful tools for studying population genetics. Nevertheless, studies of genetic diversity in stingless bees by means of these primers have revealed a low level of polymorphism, possibly the consequence of the heterologous primers used, since in most cases these were not specifically designed for the species under consideration. Herein we compared the number of polymorphic loci and alleles per locus, as well as observed heterozygosity in Melipona rufiventris and M. mondury populations, using specific and heterologous primers. The use of specific primers placed in evidence the greater frequency of polymorphic loci and alleles per locus, besides an expressive increase in observed...

Genetic divergence between populations of the stingless bee uruçu amarela (Melipona rufiventris group, Hymenoptera, Meliponini): is there a new Melipona species in the Brazilian state of Minas Gerais? Genet. Mol. Biol.
Tavares,Mara Garcia; Dias,Luiz Antonio dos Santos; Borges,Andréia Arantes; Lopes,Denilce Meneses; Busse,Angélica Helene Pereira; Costa,Ronaldo Guimarães; Salomão,Tânia Maria Fernandes; Campos,Lucio Antonio de Oliveira.
Allozyme, microsatellite and random amplified polymorphic DNA (RAPD) molecular markers were used to investigate the within and between population genetic variability and between population genetic differentiation of the Brazilian stingless bee uruçu amarela (nominally Melipona rufiventris Lepeletier, 1836) present in savanna and Atlantic forest habitats of the Brazilian state of Minas Gerais (MG). We found low levels of within population variability, although there were a large number of private alleles that specifically characterized these populations. The F ST values indicated a high level of genetic diversity between populations. Analysis of molecular variance (AMOVA) showed a high degree of population differentiation between the savanna and Atlantic...

Cytogenetic characterization of Melipona rufiventris Lepeletier 1836 and Melipona mondury Smith 1863 (Hymenoptera, Apidae) by C banding and fluorochromes staining Genet. Mol. Biol.
Lopes,Denilce Meneses; Pompolo,Sílvia das Graças; Campos,Lúcio Antônio de Oliveira; Tavares,Mara Garcia.
The stingless bees Melipona rufiventris and M. mondury were analyzed cytogenetically by conventional staining with Giemsa, C-banding and sequential staining with the fluorochromes CMA3/DA/DAPI. Both species presented 2n = 18 and n = 9, except for one colony of M. rufiventris, in which some individuals had 2n = 19 due to the presence of a B chromosome. After Giemsa staining and C-banding the chromosomes appeared very condensed and presented a high heterochromatic content, making it difficult to localize the centromere and therefore to visualize the chromosomes morphology. The constitutive heterochromatin was located in interstitial chromosome regions covering most of the chromosomes extension and consisted mainly of AT, as shown by DAPI staining. The...

Genetic variability in five populations of Partamona helleri (Hymenoptera, Apidae) from Minas Gerais State, Brazil Genet. Mol. Biol.
Borges,Andreia Arantes; Campos,Lucio Antônio de Oliveira; Salomão,Tânia Maria Fernandes; Tavares,Mara Garcia.
Partamona is a Neotropical genus of stingless bees that comprises 33 species distributed from Mexico to southern Brazil. These bees are well-adapted to anthropic environments and build their nests in several substrates. In this study, 66 colonies of Partamona helleri from five localities in the Brazilian state of Minas Gerais (São Miguel do Anta, Teixeiras, Porto Firme, Viçosa and Rio Vermelho) were analyzed using nine microsatellite loci in order to assess their genetic variability. Low levels of observed (Ho = 0.099-0.137) and expected (He = 0.128-0.145) heterozygosity were encountered and revealed discrete genetic differentiation among the populations (F ST =0.025). AMOVA further showed that most of the total genetic variation (94.24%) in P. helleri was...

Genetic divergence between Melipona quadrifasciata Lepeletier (Hymenoptera, Apidae) populations Genet. Mol. Biol.
Tavares,Mara Garcia; Pietrani,Nathalia Teixeira; Durvale,Maxwell de Castro; Resende,Helder Canto; Campos,Lucio Antonio de Oliveira.
Melipona quadrifasciata is a stingless bee widely found throughout the Brazilian territory, with two recognized subspecies, M. quadrifasciata anthidioides, that exhibits interrupted metasomal stripes, and M. quadrifasciata quadrifasciata, with continuous metasomal stripes. This study aimed to estimate the genetic variability of these subspecies. For this purpose, 127 colonies from 15 Brazilian localities were analyzed, using nine species-specific microsatellite primers. At these loci, the number of alleles ranged from three to 15 (mean: 7.2), and the observed heterozygosity (Ho) ranged from 0.03-0.21, while the expected heterozygosity (He) ranged from 0.23-0.47. The genetic distances among populations ranged from 0.03-0.45. The F ST multilocus value (0.23)...

Can climate shape flight activity patterns of Plebeia remota Hymenoptera, Apidae)? Iheringia, Sér. Zool.
Hilário,Sergio D.; Ribeiro,Márcia de F.; Imperatriz-Fonseca,Vera L..
Flight activity of foragers of four colonies of Plebeia remota (Holmberg, 1903) was registered from December 1998 to December 1999, using an automated system (photocells and PLC system). The colonies originated from two different regions: Cunha, state of São Paulo, and Prudentópolis, state of Paraná, Brazil. Flight activity was influenced by different climatic factors in each season. In the summer, the intensity of the correlations between flight activity and climatic factors was smaller than in the other seasons. During the autumn and winter, solar radiation was the factor that most influenced flight activity, while in the spring, this activity was influenced mainly by temperature. Except in the summer, the various climatic factors similarly influenced...

Comparative study of wax glands in four Meliponini bees (Hymenoptera, Apidae) producing different quantities of wax Iheringia, Sér. Zool.
Cavalcante,Vagner Melo; Oliveira,Vagner Tadeu Paes de; Cruz-Landim,Carminda da.
The developmental degree of the wax glands was compared in four Meliponini bees, that produce different quantities of wax. The histological data and height average of the wax epithelium during the time in which the maximum production of wax is expected, are in accordance with the rates of wax produced by the species. In Lestrimelitta limao (Smith, 1863) a species which has cleptobiotic habits, and frequently rob wax from the attacked colonies, the height of wax epithelium was the lowest among the studied species. The cells seem to show an abnormal vacuolated cytoplasm, in the phase in which they would be producing wax.

Protein content and electrophoretic profile of fat body and ovary extracts from workers of Melipona quadrifasciata anthidioides (Hymenoptera, Meliponini) Iheringia, Sér. Zool.
Oliveira,Vagner T. Paes de; Cruz-Landim,Carminda da.
Workers of Melipona quadrifasciata anthidioides (Lepeletier, 1836) develop their ovaries and lay eggs, therefore the production of vitellogenin is expected. In electrophoretic profiles only fat body extracts from nurse workers and ovary extracts from newly-emerged workers show protein with molecular mass similar to vitellogenin. However, an increase in the protein content was detected in forager fat body. This increase was attributed to storage of vitellogenin or other proteins in the previous phase and not discharged into the hemolymph or to an effect of the increased titre of juvenile hormone in this phase of worker life over the fat body functioning.

POST-EMBRYONIC DEVELOPMENT OF INTRAMANDIBULAR GLANDS OF FRIESELLA SCHROTTKYI (FRIESE, 1900) (HYMENOPTERA: APIDAE) WORKERS Pap. Avulsos de Zool. (São Paulo)
OLIVEIRA,ANDRÉ HENRIQUE DE; LISBOA,LUCIANE CRISTINA DE OLIVEIRA; SERRÃO,JOSÉ EDUARDO; ZANUNCIO,JOSÉ COLA; SOUZA,EDMILSON AMARAL DE.
ABSTRACT Exocrine glands play important role in social organization of insects, such as caste and inter-caste differentiation. Due their functional and structural plasticity, morphological studies on such glands contribute to better understanding the biology of social bees. Therefore, the aim of the study was to characterize the sequence of the post-embryonic development of intramandibular glands of Friesella schrottkyi (Friese, 1900) (Hymenoptera: Apidae) workers using histological and histochemical analyses. The mandibles of pupae at different developmental stages and newly emerged adults were analyzed. The intramandibular glands of F. schrottkyi presented two types: class I glands, in the mandible epidermis and class III glands, inside the mandible...

A new cluster-brood building species of Plebeia (Hymenoptera, Apidae) from eastern Brazil Rev. Bras. entomol.
Melo,Gabriel A. R.; Costa,Marco A..
A new species of Plebeia, the second largest genus of stingless bees in the Neotropical region, is described from eastern Brazil. Plebeia grapiuna sp. nov., known only from the lowland forests of southern Bahia, is most similar to P. lucii Moure, a species recently described from Minas Gerais. The lack of yellow marks and the smoother integument of the frons and mesoscutum in P. grapiuna sp. nov. distinguish them. Main features of the nesting habits of the new species are described and illustrated.
